Bile Salt Hydrolase Activities: A Novel Target to Screen Anti-Giardia Lactobacilli? ArchiMer

Giardia duodenalis is a protozoan parasite responsible for giardiasis, a disease characterized by intestinal malabsorption, diarrhea and abdominal pain in a large number of mammal species. Giardiasis is one of the most common intestinal parasitic diseases in the world and thus a high veterinary, and public health concern. It is wellestablished that some probiotic bacteria may confer protection against this parasite in vitro and in vivo and we recently documented the implication of bile-salt hydrolase (BSH)-like activities from strain La1 of Lactobacillus johnsonii as mediators of these effects in vitro. We showed that these activities were able to generate deconjugated bile salts that were toxic to the parasite. In the present study, a wide collection of...

Diagnosis of Giardia infections by PCR-based methods in children of an endemic area J. Venom. Anim. Toxins incl. Trop. Dis.
David,EB; Coradi,ST; Oliveira-Sequeira,TCG; Ribolla,PEM; Katagiri,S; Guimarães,S.
The present study was designed to estimate the prevalence of Giardia infection in preschool- and school-aged children living in an endemic area. Fecal samples from 573 children were processed by zinc sulfate centrifugal flotation, centrifugal sedimentation (using a commercial device for fecal concentration - TF-Test kit®) and polymerase chain reaction (PCR)-based methods. Of the stool samples assessed, 277 (48.3%) were positive for intestinal parasites and/or commensal protozoa. Centrifugal flotation presented the highest diagnostic sensitivity for Giardia infections. The kappa index revealed that both coproparasitological techniques closely agreed on the Giardia diagnosis (86%) versus satisfactory (72%) and poor (35%) concordances for commensal protozoan...

Genotyping of Brazilian Giardia duodenalis human axenic isolates J. Venom. Anim. Toxins incl. Trop. Dis.
Coradi,ST; David,EB; Oliveira-Sequeira,TCG; Ribolla,PEM; Carvalho,TB; Guimarães,S.
Giardia duodenalis is a complex species that comprises at least seven distinct genetic groups (A to G), but only genotypes A and B are known to infect humans and a wide variety of other mammals. Regardless of biological, biochemical and antigenic analysis, several isolates maintained in vitro were not genetically typed yet. So, in the present study, five Brazilian axenic isolates obtained from asymptomatic and symptomatic patients were typed in order to determine the major genetic groups to which the isolates belonged. DNA was extracted from axenic trophozoites, fragments of glutamate dehydrogenase (gdh) and triosephosphate isomerase (tpi) genes were amplified by PCR and the isolate genotyping was carried out using restriction fragment length polymorphism...

Giardiasis as zoonosis: between proof of principle and paradigm in the Northwestern region of São Paulo State, Brazil BJID
Volotão,Aline Cardoso Caseca; Ramos,Nathália Motta Delvaux; Fantinatti,Maria; Moraes,Marcus Vinícius Proença de; Atique Netto,Halim; Storti-Melo,Luciane Moreno; Godoy,Elenir Alves Macedo de; Rossit,Andréa Regina Baptista; Fernandes,Octávio; Machado,Ricardo Luiz Dantas.
OBJECTIVE: In order to evaluate the potential zoonotic transmission of Giardia duodenalis, isolates from humans and dogs in the Northwestern region of the São Paulo State, Brazil were characterized based on the β-giardin gene. METHODS: The samples were analyzed by sequencing of the Nested-PCR products. RESULTS: The A1 and A2 subgenotypes were detected in human and dogs. Cysts of assemblage B, C and D have not been found in any isolates studied. CONCLUSIONS: These results are consistent with the view that giardiasis in the largest endemic region of the Brazil should not be seen as a single entity.

Modification of the Alere GIARDIA Ag TEST immunochromatography KIT methodology for its use in frozen fecal sediment of dogs and cats Anais da ABC (AABC)
COSTA,VIVIANE A.N.; BRENER,BEATRIZ; FONSECA,ANA BEATRIZ M.; SUDRÉ,ADRIANA P..
ABSTRACT Giardia duodenalis is a worldwide intestinal parasite and is one of the most frequent protozoa species infecting dogs and cats. This study aimed to modify the methodology of Alere GIARDIA Ag TEST KIT for its use in frozen fecal sediments with different storage times in a freezer (-20°C), thus expanding the range of use of this methodology. One hundred fecal sediments from dogs (n=50) and cats (n=50) previously examined by optical microscopy for Giardia cysts were selected for this study. The agreement between the modified immunochromatography and microscopy results was calculated by Kappa coefficient. To evaluate the performance of the modified immunochromatography assay on samples with different storage time, the fecal sediments were divided into...

Remoción de formas parasitarias intestinales en una laguna facultativa de estabilización en Lima, Perú Zoologia
Iannacone,José O..
The effectiveness of a waste stabilization pond, to remove intestinal helminth eggs and protozoan cysts was studied in San Martin de Porres, Lima, Peru. From September to October 2000 four collections of samples were performed from raw domestic wastewater and from contends of primary, secondary and tertiary ponds. A comparative study of performance of the concentrations methods of Ritchie and Faust was also done. The overall average removal of parasitic forms was as follow: 69.37% from raw water and primary pond; 48.34% from primary to secondary pond; 85.45% from secondary to tertiary pond and 97.69% from raw water to tertiary pond. There was no significant difference in the quantitative performance of Ritchic and Faust methods. Giardia duodenalis (Lambl,...
